Abstract
Acclimatisation as the final stage in micropropagation was investigated owing to its importance in the survival of 'in vitro' grown plants, Differences between the 'in vitro' and 'ex vitro' environment result in physiological and morphological abnormalities, reflecting the way in which plantlets adapt to in vitro conditions. Variations in relative humidity, light intensity and sucrose concentration in 'in vitro' grown Rosa sp. cv. Red cascade were examined in relation to wax content, stomatal density, chlorophyll content and consequent plantlets survival rate. In conjunction with low sucrose concentration, a gradual reduction in relative humidity 'in vitro' to 86% resulted in a 100% survival rate of plantlets directly transferred into pots. Reducing relative humidity during the rooting stage reduced acclimatisation duration. Survival rate was decreased as the light intensity decreased. Overall results indicate that cultured plantlets can resemble greenhouse-grown plants under modified environmental conditions, thus increasing the economic potential of large-scale, commercial tissue culture ventures.
